Nexium (esomeprazole) contraindications and overdosage

Esomeprazole magnesium - Nexium belongs to the class of drugs known as proton pump inhibitors (PPIs). PPIs are used to treat medical conditions such as stomach ulcers and gastroesophageal reflux disease - GERD, reflux esophagitis by reducing the amount of acid the stomach produces. Nexium is also used along with certain antibiotics to treat stomach ulcers that are caused by bacteria known as H. pylori.

Nexium contraindications

Do not use Nexium if you are allergic to esomeprazole or to any other benzimidazole medication such as albendazole or mebendazole. Tell your healthcare provider if you have ever had liver disease or any type of heart disease, before taking Nexium. You may need a dose adjustment or special tests during treatment with Nexium.

Some medical conditions are treated with a combination of esomeprazole and antibiotics. To best treat your condition, use all of your medications as prescribed by your healthcare provider. Do not change your doses or medication schedule without advice from your healthcare provider.

Nexium overdosage

The major signs of acute toxicity with Nexium were reduced motor activity, changes in respiratory frequency, ataxia, tremor and intermittent clonic convulsions. Some overdosage cases with esomeprazole were reported. Reports have been received of overdosage with omeprazole (chemicaly very similar to esomeprazole) in humans. Doses ranged up to 2,400 mg which is 120 times the usual recommended dose. Side effects were variable, but included tachycardia, confusion, diaphoresis, drowsiness, blurred vision, flushing, headache, dry mouth, nausea and other adverse reactions similar when normal clinical dosages are taken.

There is no specific antidote for esomeprazole and since esomeprazole is extensively protein bound, it is not expected to be removed by dialysis. In case of overdosage, treatment should be symptomatic and supportive. As with the management of any overdose, the possibility of multiple medication ingestion should be considered.
